The CellTiter 96® AQueous One Solution is a colorimetric cell proliferation assay based on the conversion of an MTS tetrazolium compound to a colored formazan product by viable cells. The MTS reduction is accomplished by NADPH or NADH produced by dehydrogenase enzymes in metabolically active cells.

The absorbance of the formazan at 490nm can be measured directly from 96-well assay plates without additional processing. The conversion of MTS into aqueous, soluble formazan is accomplished by dehydrogenase enzymes found in metabolically active cells.

PES has enhanced chemical stability, which allows it to be combined with MTS to form a stable solution. This convenient “One Solution” format is an improvement over the first version of the CellTiter 96® AQ Assay, where phenazine methosulfate (PMS) is used as the electron.
