The Configuration of GRB2 in Protein Interaction and Signal ...
Growth-factor-receptor-binding protein 2 (GRB2) is a non-enzymatic adaptor protein that plays a pivotal role in precisely regulated signaling cascades from cell surface receptors to cellular responses, including signaling transduction and gene expression.
LRRK1 forms a complex with EGFR via Grb2 in response to EGF ...
Download scientific diagram | LRRK1 forms a complex with EGFR via Grb2 in response to EGF stimulation.(a) Interaction of LRRK1 with Grb2. COS7 cells were co-transfected with Flag-LRRK1, Flag-LRRK2 ...
LRRK2 and its substrate Rab GTPases are sequentially targeted …
Sep 12, 2018 · Here, we reveal a key stress-responsive pathway composed of Rab7L1, LRRK2, and phosphorylated Rab8/10 involved in lysosomal homeostasis. Lysosomal overload stress induces translocation of Rab7L1 and LRRK2 to lysosomes, where LRRK2 is activated, and stabilizes Rab8 and Rab10 through phosphorylation.
Leucine-rich repeat kinase 2 at a glance - PMC
Leucine-rich repeat kinase 2 (LRRK2) is a multidomain scaffolding protein with dual guanosine triphosphatase (GTPase) and kinase enzymatic activities, providing this protein with the capacity to regulate a multitude of signalling pathways and act as a …
Grb2, a Double-Edged Sword of Receptor Tyrosine Kinase Signaling
Grb2 is conventionally known for playing positive roles in RTK signaling. The discovery of a negative regulatory role for Grb2 reveals that this adaptor acts as a double-edged sword in the regulation of RTK signaling.
GRB2: A dynamic adaptor protein orchestrating cellular signaling …
Sep 1, 2024 · GRB2, or Growth Factor Receptor-Bound Protein 2, is a pivotal adaptor protein in intracellular signal transduction pathways, particularly within receptor tyrosine kinase (RTK) signaling cascades.
Ataxin-2 Modulates the Levels of Grb2 and Src but Not Ras …
Jan 19, 2013 · Our present data show for the first time that ATXN2 indeed interacts with the SH3 motif containing protein Grb2 in the endocytosis machinery and that ATXN2 loss-of-function and gain-of-function mutations in fibroblasts modulate Grb2 protein levels.
LRRK2 and its substrate Rab GTPases are sequentially targeted …
Sep 25, 2018 · Here, we reveal a mechanism of stress-induced lysosomal response by LRRK2 and its target Rab GTPases. Lysosomal overload stress induced the recruitment of endogenous LRRK2 onto lysosomal membranes and activated LRRK2. An upstream adaptor Rab7L1 (Rab29) promoted the lysosomal recruitment of LRRK2.
Structural biology of the LRRK2 GTPase and kinase domains: …
May 5, 2014 · In this review we highlight the recent progress in structural and biochemical characterization of Roco proteins and discuss its implication for the understanding of the complex regulatory mechanism of LRRK2. Keywords: LRRK2, Roco, structure, kinase, G-protein, Parkinson’s disease.
GRB2 - Wikipedia
Grb2 is best known for its ability to link the epidermal growth factor receptor tyrosine kinase to the activation of Ras and its downstream kinases, ERK1,2. Grb2 is composed of an SH2 domain flanked on each side by an SH3 domain. Grb2 has two closely related proteins with similar domain organizations, Gads and Grap.
