CCL2 and CXCL8 enhance neutrophil chemotactic ability in cells from patients with RA, and CCR2 KO mice are resistant to AIA model through the suppression of CCL2/CCR2-induced neutrophil recruitment . CCL3 expression is associated with the neutrophil number in the SFs from patients with RA ( 106 ).

These data provide evidence that the chemokine signaling pathway is involved in CCL2 expression in RA patient tissues, which may contribute to chronic inflammation associated with RA. Targeting this signaling pathway may provide a novel therapeutic avenue in RA.

CCL2 increases VCAM-1 production by binding to CCR2 receptor, and activating PKCδ and p38 which in turn enhances binding of AP-1, resulting in the transactivation of VCAM-1 expression. The CCL2-mediated CCR2, PKCδ, p38MAPK, and AP-1 pathway promotes monocytes adhesion to human OASFs.

This current study found significantly higher levels of CCN1 and CCL2 expression in RA synovial fluid compared with non-RA synovial fluid. Moreover, we identified CCL2 as a target protein for the CCN1 signaling pathway regulating monocyte infiltration.
