The sgRNA is formed by the fusion of crRNA and tracrRNA with engineered tetra loop structure (green). The sgRNA sequence consists of four different sequences, spacer (blue), crRNA repeat (brown), tetra loop (green) and tracrRNA (purple). It also has six structural secondary structure, spacer, lower stem, bulge, upper stem, nexus and hairpins.
How to design sgRNA sequences - Takara Bio
CRISPR/Cas9 gene targeting requires a custom single guide RNA (sgRNA) that contains a targeting sequence (crRNA sequence) and a Cas9 nuclease-recruiting sequence (tracrRNA). The crRNA region (shown in red below) is a 20-nucleotide sequence that is homologous to a region in your gene of interest and will direct Cas9 nuclease activity.

Guide RNA - Wikipedia
Guide RNA (gRNA) or single guide RNA (sgRNA) is a short sequence of RNA that functions as a guide for the Cas9-endonuclease or other Cas-proteins [1] that cut the double-stranded DNA and thereby can be used for gene editing. [2]

CRISPR Guide - Addgene
Engineered CRISPR systems contain two components: a guide RNA (gRNA or sgRNA) and a CRISPR-associated endonuclease (Cas enzyme). The gRNA is a short synthetic RNA composed of a scaffold sequence necessary for Cas-binding and a user-defined ∼20-nucleotide spacer that defines the genomic target to be modified.

Diagram showing 11 sgRNA sites targeting albino exons 1 and 2 used in an independent validation of the prediction model. Phenotypes obtained after the injection of the sgRNAs, showing different levels of mosaicism compared to the wild type (WT) (bottom).
Sequence determinants of improved CRISPR sgRNA design
We systematically assessed the DNA sequence features that contribute to single guide RNA (sgRNA) efficiency in CRISPR-based screens. Leveraging the information from multiple designs, we derived a new sequence model for predicting sgRNA efficiency in …

Jul 2, 2019 · Which is better, two-part (tracrRNA/crRNA) or single guide RNA (sgRNA)? Some reports have stated that single guide RNAs outperform two-part guide RNAs (tracrRNA and a crRNA) in most or all cases. Here, we compare the two types of guide RNA complexes directly.
