Crosstalk between Bcl-2 family and Ras family small GTPases: …
Here we attempt to present a brief review of the crosstalk between the Bcl-2 family members and the small guanosine triphosphatases (GTPases) of the Ras superfamily, which also modulate the pro- and anti-apoptotic properties of Bcl-2 members.
Regulation of Bcl-2 gene expression by BCR-ABL is mediated by Ras
Mar 28, 1997 · The data presented here establish that Bcl-2 is a downstream target gene of the Ras signalling pathway in cells transformed by BCR-ABL, and that constitutive Ras activation results in constitutive expression of the gene.
Bcl-2, via Its BH4 Domain, Blocks Apoptotic Signaling Mediated by ...
Here, we demonstrate that, upon apoptotic stimulation, newly synthesized Bcl-2 redistributes to mitochondria, interacts there with activated Ras, and blocks Ras-mediated apoptotic signaling. We also show, by employing bcl-2 mutants, that the BH4 domain of Bcl-2 binds to Ras and regulates its anti-apoptotic activity.
Targeting Bcl-2 stability to sensitize cells harboring oncogenic ras
Sep 8, 2015 · Studies are shown that cells harboring an oncogenic ras were extremely sensitive to the inhibition of protein kinase C (PKC) and Bcl-2 could antagonize this apoptotic process. However, it remains unrevealed how Bcl-2 is being regulated in this apoptotic process.
Bcl-2, via its BH4 domain, blocks apoptotic signaling mediated by ...
Feb 21, 2003 · Experiments with a C-terminal-truncated Ras or a farnesyltransferase inhibitor demonstrate that the CAAX motif of Ras is essential for apoptotic signaling and Bcl-2 association. The results indicate a potential mechanism by which Bcl-2 protects cells against Ras-mediated apoptotic signaling.
Crosstalk between Bcl-2 family and Ras family small GTPases ... - PubMed
Jan 2, 2013 · Similar functional associations have been described between the Bcl-2 family and other members of the Ras superfamily. These interactions at the mitochondria provide novel opportunities for strategic therapeutic targeting of drug-resistant cancers.
Bcl-2 associates with the ras-related protein R-rasp23 - Nature
Nov 18, 1993 · Overexpression of the proto-oncogene bcl -2 can override these signals and block apoptosis 4–14, indicating that the bcl-2 protein (Bcl-2) is an important component of the apoptotic response....
Bcl-2 promotes premature senescence induced by oncogenic Ras
Apr 11, 2003 · Here we show that constitutive expression of Bcl-2 accelerates selected features of the Ras-induced senescence program in primary human fibroblasts. Yet, Bcl-2 also inhibits fibroblast apoptosis induced by exogenous H 2 O 2, while both signals induce an increased endogenous Bcl-2 expression in these cells.
BCL-2 family isoforms in apoptosis and cancer - Nature
Feb 21, 2019 · BCL-2 family members play an integral role in apoptosis, but also contribute to many other cellular functions. Isoforms of almost all of the BCL-2 family members have been identified and some are...
