A function of p21 during promyelocytic leukemia cell ... - PubMed
May 27, 1999 · Differentiation is mediated by the APL-specific transforming protein PML/RAR alpha and involves its activity as ligand-dependent enhancer factor on RA-target genes. We report here the identification of p21 as a transcriptional target of PML/RAR alpha during RA-induced differentiation of APL cells.
PML Is a Direct p53 Target that Modulates p53 Effector Functions
Feb 27, 2004 · Previous work suggests that the promyelocytic leukemia gene (PML) can act upstream of p53 to enhance transcription of p53 targets by recruiting p53 to nuclear bodies (NBs). We show that PML is itself a p53 target gene that also acts downstream of p53 to potentiate its antiproliferative effects.
PML restrains p53 activity and cellular senescence in clear cell …
May 10, 2024 · Here, we report a novel ccRCC dependence on the promyelocytic leukemia (PML) protein. We show that PML is overexpressed in ccRCC and that PML depletion inhibits cell proliferation and relieves pathologic features of anaplastic disease in vivo.
The Role of PML in Tumor Suppression - Cell Press
Jan 25, 2002 · The PML gene, involved in the t(15;17) chromosomal translocation of acute promyelocytic leukemia (APL), encodes a protein which localizes to the PML-nuclear body, a subnuclear macromolecular structure. PML controls apoptosis, cell proliferation, and senescence. Here, we review the current understanding of its role in tumor suppression.
A function of p21 during promyelocytic leukemia cell ... - Nature
May 25, 1999 · We report here the identification of p21 as a transcriptional target of PML/RARα during RA-induced differentiation of APL cells. We found that RA-treated APL cells undergo two rounds of cell...
Extracellular-regulated protein kinase 5-mediated control of p21 ...
Aug 8, 2020 · Based on previous analyses of ERK5 in cancer cells, ERK5 activity may promote macrophage proliferation by suppressing p53-mediated transcriptional upregulation of p21 through blocking PML-dependent sequestration of MDM2 in cancer cells (38, 39).
MOZ increases p53 acetylation and premature senescence …
Mar 5, 2013 · As a result of PML-mediated suppression of Akt, the increased PML-MOZ interaction enhances p21 expression and induces p53-dependent premature senescence upon forced PML expression. Our research demonstrates that MOZ controls p53 acetylation and transcriptional activity via association with PML.
yelocytic leukemia protein (PML) contributes to the up-regulation of p21 in a p53-independent pathway. Knock-down of PML in p53-null H1299 and HCT 116 (p53–/–) tumor cells by specific siRNA resulted in down-regulation of p21 protein expres.
Increased p21 in PML/RARa cells during G1-S had no e ect on the cell cycle while an antisense p21 prevented RA-induced di erentiation without altering G1-S transition and the late G1 block.
The function of PML in p53-dependent apoptosis - PubMed
The PML gene of acute promyelocytic leukaemia (APL) encodes a growth- and tumour-suppresor protein that is essential for several apoptotic signals. The mechanisms by which PML exerts its pro-apoptotic function are still unknown. Here we show that PML acts as a transcriptional co-activator with p53. …
