Lysosomal-associated membrane protein 1 (LAMP-1) also known as lysosome-associated membrane glycoprotein 1 and CD107a (C luster of D ifferentiation 107a), is a protein that in humans is encoded by the LAMP1 gene. The human LAMP1 gene is located on the long arm (q) of chromosome 13 at region 3, band 4 (13q34).

Jul 20, 2023 · Here, we use functional proteomics, single-particle cryo-EM, electrophysiology, and in vivo imaging to unravel a key biological function of human lysosome-associated membrane proteins (LAMP-1 and LAMP-2) in regulating lysosomal pH homeostasis.

Their structure is characterized by a highly glycosylated luminal region forming a glycoprotein layer in the lysosomal lumen, a transmembrane region, and a short C-terminal cytosolic domain (Fig. 1). LAMP1 has only one transcript, whereas LAMP2 has three different splicing isoforms: LAMP2A, LAMP2B, and LAMP2C [30,33].

Despite widespread distribution patterns of LAMP1 and the heterogeneous nature of LAMP1-labeled compartments, LAMP1 continues to be the most routinely used marker for neuronal lysosomes, and many studies refer to LAMP1-positive organelles as degradative lysosomes.

The lysosomal membrane proteins LAMP-1 and LAMP-2 are estimated to contribute to about 50% of all proteins of the lysosome membrane. Surprisingly, mice deficient in either LAMP-1 or LAMP-2 are viable and fertile. However, mice deficient in both LAMP-1 and LAMP-2 have an embryonic lethal phenotype.
