Upon activation with LPS, almost 90% of all NF-κB is localized in the nucleus of macrophages (Fig. 4A, whereas only 60% of NF-κB was localized in the nucleus of the RAW 264.7 cell line (Fig. 4B). Mathematical modeling revealed that such fast activation could be regulated by the strength of IKK complex activation.

In summary, our data indicate that NF-κB activation by LPS is not critically involved in disruption of endothelial barrier properties. Rather, our data suggest that NF-κB activation acts as a part of a rescue mechanism. Sepsis is one of the leading causes of death in critically ill patients (Levy et al. 2010; Russell 2006).

Post-LPS activation of NF-κB phosphorylation peaked at 5 min, whereas IκBα degradation and NF-κB nuclear translocation kinetics peaked at 10 min. Signaling kinetics were substantially faster in comparison to the transformed RAW 264.7 macrophage cell line 8, where NF-κB nuclear translocation peaked at 30 min after LPS stimulation.
