HLA-DR5 (DR5) is a broad- antigen serotype that is further split into HLA-DR11 and HLA-DR12 antigen serotypes. Except for a few DRB1* alleles, the reactivity of DR5 is relatively poor.

The HLA-DRB4 locus encodes the HLA-DR53 specificity, has some variation, and is associated with certain HLA-DRB1 types. The HLA-DRB5 locus encodes the HLA-DR51 specificity, which is typically invariable, and is linked to the HLA-DR2 types.

In this study, the HLA-DR5 allele was associated with development of a defined connective tissue disease, while HLA-DR2 or DR4 was more likely associated with nondifferentiation into a defined connective tissue disease.

In this chapter, we focus on the state-of-the-art classification of HLA supertypes including HLA-I supertypes and HLA-II supertypes and their application in development of peptide-based vaccines. Key words: Peptide, Vaccine, HLA-I supertypes, HLA-II supertypes.
