In this study, we show that HDAC1 can be both neuroprotective and neurotoxic depending on whether it interacts with the Class IIA HDAC, HDRP, or with HDAC3, a member of the Class I family. The neurotoxic effect of HDAC1 is dependent on GSK3β activity and can be inhibited by IGF-1 treatment or activation of PI3K-Akt signaling.
