ETV1, 4 and 5: An Oncogenic Subfamily of ETS Transcription Factors
Here, we review evidence for a role of ETV1, 4 and 5 as oncoproteins and describe modes of their action. Modulation of their activation or interaction with cofactors as well as inhibiting crucial target gene products may ultimately be exploited to treat various cancers that are dependent on the PEA3 group of ETS transcription factors.
Function and regulation of the PEA3 subfamily of ETS transcription ...
Structures and functions of the PEA3 subfamily ETV1, ETV4, and ETV5 are protein-coding genes with DNA-binding transcription factor activity. In humans, ETV1 is located on chromosome 7q22, whereas ETV4 and ETV5 are located on 17q21 and 3q27, respectively.
ETV1 Gene - GeneCards | ETV1 Protein | ETV1 Antibody
Mar 30, 2025 · ETV1 (ETS Variant Transcription Factor 1) is a Protein Coding gene. Diseases associated with ETV1 include Ewing Sarcoma and Soft Tissue Sarcoma. Among its related pathways are Akt Signaling and MAPK-Erk Pathway.
Transcriptional regulation of ETV5 by mitogen-activated ... - Nature
Apr 9, 2025 · In cancer tissues, ETV1, ETV4, and ETV5 are highly expressed and their biological functions are synergistic 49. ETV4 and ETV5 play a compensatory role each other and necessary for efficient ...
Relationship between ETS Transcription Factor ETV1 and TGF-β
Jun 3, 2019 · In this report, we identified SMAD4 as a novel interaction partner of ETV1 that can repress ETV1-mediated transcription, providing a mechanism by which TGF-β signaling may constrain ETV1’s ...
Structures of the Ets Protein DNA-binding Domains of …
May 5, 2015 · Here, we present the crystal structures of the Ets domains of the entire PEA3 subfamily (Etv1, Etv4, and Etv5) together with the structure of the Erg family member (Fev). All proteins share a highly conserved core Ets structure and make extensive interactions with DNA.
Structures of the Ets Protein DNA-binding Domains of ... - PubMed
May 29, 2015 · Homodimers of Etv1, Etv4, and Etv5 could be reduced to monomers, leading to a 40-200-fold increase in DNA binding affinity. Hence, we present the first indication of a redox-dependent regulatory mechanism that may control the activity of this subset of oncogenic Ets transcription factors.
Transcription factor Etv5 is essential for the maintenance of …
How vertebrate CRL4 COP1/DET1 is regulated is less clear. Here we show that Etv5 is expressed specifically in AT2 cells and is stabilized when CRL4 COP1/DET1 is inhibited by ERK signaling. Genetic deletion of Etv5 in AT2 cells revealed that Etv5 is …
The ETS family of oncogenic transcription factors in solid tumours
Apr 28, 2017 · ETS factor expression is aberrantly upregulated in solid tumours through chromosomal translocation and amplification. Activating mutations in KIT stabilize the ETV1 protein through the MEK–ERK...
ETV1, 4 and 5: an oncogenic subfamily of ETS transcription factors
The homologous ETV1, ETV4 and ETV5 proteins form the PEA3 subfamily of ETS transcription factors. In Ewing tumors, chromosomal translocations affecting ETV1 or ETV4 are an underlying cause of carcinogenesis.
