E2F1: Cause and Consequence of DNA Replication Stress
The study, conducted in our laboratory, shows that E2F1 is degraded upon treatment with Chk1 inhibitors, and that the E3 ligase cyclin F is the main mediator of this proteasomal degradation (Burdova et al., 2019).
Methylated DNMT1 and E2F1 are targeted for proteolysis by
Apr 24, 2018 · DNMT1 contains a consensus methylation motif shared by many non-histone proteins including E2F1, a key transcription factor for S phase. We show that the methylation-dependent E2F1 degradation...
Regulation of E2F1 Transcription Factor by Ubiquitin Conjugation
E2F1 harbors 14 lysine residues that are all potential acceptors for ubiquitin modification. E2F1’s abundance fluctuates along the cell cycle and in response to DNA damage, but also in response to differentiating agents.
Calcineurin-mediated dephosphorylation stabilizes E2F1 protein …
Oct 4, 2024 · We here identified SCF FBXW7 as a ubiquitin ligase for E2F1 by comprehensive analysis. We found that phosphorylation of E2F1 at serine-403 promotes its binding to FBXW7 (F-box/WD repeat-containing protein 7) followed by its ubiquitination and degradation.
Structural mechanism for recognition of E2F1 by the ubiquitin ligase ...
Jan 16, 2025 · We utilized single-particle cryo-electron microscopy to elucidate the structure of a Cyclin F-Skp1 complex bound to an E2F1 peptide. The structure and biochemical analysis reveal important differences in the substrate-binding site of Cyclin F compared to Cyclin A.
Structural mechanism for recognition of E2F1 by the ubiquitin ligase ...
Jan 15, 2025 · Unlike canonical cyclins, which bind and activate cyclin-dependent kinases (CDKs), Cyclin F functions as a substrate receptor protein within the Skp1-Cullin-F box (SCF) E3 ubiquitin ligase complex, enabling the ubiquitylation of target proteins.
DNA damage and S phase-dependent E2F1 stabilization requires …
May 25, 2017 · Here, we demonstrated that the E3-ubiquitin ligase cellular inhibitor of apoptosis 1 (cIAP1) increases E2F1 K63-poly-ubiquitination on the lysine residue 161/164 cluster, which is associated with...
Here, we highlight recent studies that implicate the Skp1-Cul1-F-box-protein (SCF) ubiquitin ligase family as a key factor in the control of the E2F family of cell cycle transcriptional regulators.
DNA damage and S phase-dependent E2F1 stabilization requires ... - PubMed
May 25, 2017 · The E2F transcription factor 1 is subtly regulated along the cell cycle progression and in response to DNA damage by post-translational modifications. Here, we demonstrated that the E3-ubiquitin ligase cellular inhibitor of apoptosis 1 (cIAP1) increases E2F1 K63-poly-ubiquitination on the lysine res …
Regulation of E2F1 by APC/CCdh1 via K11 linkage-specific …
May 5, 2012 · Our studies show that APC/C ubiquitin ligase in conjunction with its co-activator Cdh1 (APC/C Cdh1) can downregulate E2F1. We also identify an APC/C subunit APC5 that binds to E2F1 and is essential for E2F1 ubiquitination. We confirm an interaction between E2F1 and Cdh1 as well as an interaction between E2F1 and APC5 both in vivo and in vitro.
