CARD8: A Novel Inflammasome Sensor with Well-Known Anti …
The CARD8 T60 isoform was identified as a sensor for HIV protease activity in HIV-1-infected CD4 + T cells and macrophages . Pathogen sensing by CARD8 resulted in pyroptosis of infected cells and IL-1β secretion in macrophages, THP-1 cells, or transfected HEK293 cells [ 62 ].
The CARD8 T60 variant associates with NLRP1 and negatively …
Nov 7, 2022 · In this study, we identified that the “canonical” T48 isoform of CARD8 interacts with human inflammasome sensor NLRP1, but fails to suppress its activation. Surprisingly, we found the somewhat larger T60 isoform of CARD8 could act as a negative regulator to control the NLRP1 inflammasome activation.
Activation of the CARD8 Inflammasome Requires a Disordered Region
Oct 13, 2020 · To investigate the function of the putative disordered region of CARD8, we transiently transfected constructs encoding either full-length CARD8 (isoform 5, the longest isoform) or the ZU5-UPA-CARD (ZUC) domains of CARD8 (starting at residue 162) into HEK293T cells stably expressing CASP1 and GSDMD.
Loss-of-function CARD8 mutation causes NLRP3 inflammasome …
May 1, 2018 · Our studies show that a somewhat larger isoform of CARD8, the T60 isoform having a start site in exon 4, also binds to NLRP3 and inhibits its activity.
Loss-of-function CARD8 mutation causes NLRP3 inflammasome …
May 1, 2018 · Whole exome sequencing and PCR studies identified the affected individuals as having a V44I mutation in a single allele of the T60 isoform of CARD8.
Novel isoforms of the CARD8 (TUCAN) gene evade a nonsense …
CARD8 (TUCAN) is implicated in the regulation of apoptosis and inflammation, and is a positional and functional candidate gene for inflammatory bowel disease (IBD). Recent investigations have reported conflicting results of association between a …
CARD8: A Novel Inflammasome Sensor with Well-Known Anti
Jun 13, 2024 · Alternative splicing leads to the generation of multiple CARD8 protein isoforms. Although the functional properties of these isoforms are poorly characterized, there is evidence that suggests isoform-specific roles.
