BRD4 - Wikipedia
Bromodomain-containing protein 4 is a protein that in humans is encoded by the BRD4 gene. [5] [6] BRD4 is a member of the BET (bromodomain and extra terminal domain) family, which also includes BRD2, BRD3, and BRDT. [7] BRD4, similar to other BET family members, contains two bromodomains that recognize acetylated lysine residues. [8]

BRD4 and Cancer: going beyond transcriptional regulation
Nov 22, 2018 · BRD4, member of the Bromodomain and Extraterminal (BET) protein family, is largely acknowledged in cancer for its role in super-enhancers (SEs) organization and oncogenes expression regulation.
Inhibition of BRD4 shortcuts the communication between SEs and target promoters with a subsequent cell-spe …
Emerging roles of and therapeutic strategies targeting BRD4 in …
These findings provide insight into the role of BRD4 in epigenetic regulation of gene pathways that promote the metastatic phenotype of cancer cells and suggest that therapies targeting BRD4 may represent a strategy to prevent cancer metastasis.

Dec 22, 2023 · Small molecule compound BRD4 BD1 and BD2 bromodomain inhibitors block BRD4 binding to super-enhancers, suppress oncogene transcription and expression, reduce cancer cell proliferation and...
BRD4: a general regulator of transcription elongation - PMC
BRD4 localizes to the promoter-proximal region of genes. At the target gene BRD4 helps to assemble a functional elongation complex at the 5’-end of genes. During this general 5’-elongation control point BRD4 also recruits 3’-end RNA processing factors.
Regulation of programmed cell death by Brd4 | Cell Death
Dec 20, 2022 · Epigenetic factor Brd4 has emerged as a key regulator of cancer cell proliferation. Targeted inhibition of Brd4 suppresses growth and induces apoptosis of various cancer cells.
SMARCA4 regulates inducible BRD4 genomic redistribution …
Mar 25, 2025 · The BRD4 complex coordinate key regulatory steps in gene expression. We speculate on why SMARCA4 recruits BRD4 to cell stress and motility pathways. BRD4 is a pleiotropic protein most intensively studied as a regulator of transcriptional elongation, a highly regulated step in expression of intrinsic innate genes.
Distinct layers of BRD4-PTEFb reveal bromodomain ... - Cell Press
Aug 17, 2023 · Although BRD4-PTEFb can associate with chromatin through acetyl recognition, our results indicate that a distinct, active BRD4-PTEFb population functions to regulate transcription independently of bromodomain-mediated chromatin association. These findings may enable more effective pharmaceutical modulation of BRD4-PTEFb activity.
