Extrapolating AUC to Infinity - Certara
The extrapolation of AUC to infinity requires several assumptions. The first assumption is that after the last measurable concentration, drug declines in mono-exponential fashion. The second assumption is that the elimination rate constant of that mono-exponential decline is accurately estimated by the terminal elimination rate constant ...
Pharmacokinetics – Area Under the Curve Calculations
The area under the concentration vs. time curve is a useful description of exposure of drug following administration. There are different ways in which AUC may be calculated, and terminology may vary from one source or text to another. This page summarizes the calculations and terminology that may be encountered.
Area under the curve (pharmacokinetics) - Wikipedia
In the field of pharmacokinetics, the area under the curve (AUC) is the definite integral of the concentration of a drug in blood plasma as a function of time (this can be done using liquid chromatography–mass spectrometry [1]).
AUC is approximated by a series of trapezoids. Compute the area of all trapezoids and sum them to give the AUC up to the last sample drawn. AUC(0-inf): AUC curve to infinite time. As we cannot get the assays to go to infinity, we have to extrapolate to infinity.
Assessment of Pharmacologic Area Under the Curve When …
In pharmacology, the area under the plot of plasma concentration of a drug versus time after dosage (called “area under the curve” or AUC) gives insight into the extent of exposure to a drug and its clearance rate from the body.
AUC in Pharmacokinetics - PharmaEducation
Nov 1, 2024 · AUC in pharmacokinetics (Area Under the Curve) is a vital parameter of the plasma level time curve. AUC is also known as the zero moment curve. It is a crucial measure of the bioavailability of a drug after administration.
Pharmacokinetic terms: symbols and units - Wiley Online Library
Dec 21, 2001 · Area under the curve (AUC) is expressed in units of μg · h/mL (μg × h/mL) AUC total area under the plasma drug concentration–time curve (from time zero to infinity).
NCA parameter formulas - Certara
Jul 9, 2020 · AUCall: Area under the curve from the time of dosing to the time of the last observation. If the last concentration is positive, AUClast=AUCall. Otherwise, AUCall will not be equal to AUClast, as it includes the additional area from the last measurable (positive) concentration down to zero or negative observations.

Mar 4, 2024 · Objective: In pharmacokinetics, the area under the concentration versus time curve (AUC) extrapolated to infinity (AUC0-∞) is the preferred metric but it is not always possible to have a reliable estimate of the terminal phase half-life.
